Description

SOD is an important antioxidant enzyme that catalyzes the dismutation of superoxide anion, (which is among the reactive oxygen species that are produced from the metabolism of various cells including free radicals), to oxygen and hydrogen peroxide, and has been used as an indicator to measure antioxidant capacity. XO/NBT has been a widely used method for the assay of SOD, but NBT forms formazan, which is not soluble in water, and reacts directly with xanthine oxidase, making accurate measurement difficult . OxiTec™ SOD Assay kit is able to measure more stable and accurate SOD activity using Water Soluble Tetrazolium salt (WST) to compensate for these problems Xanthine oxidase catalyzes conversion of hypoxanthine and xanthine into uric acid, and uses oxygen as redox partner, producing ROS such as and H2O2 and O2-. This O2- product reduces WST-1 to WST-formazan, which exhibits absorbance at 450 nm.
However, when SOD is active, superoxide ion levels are reduced, inhibiting the formation of WST-formazan. The degree of inhibition is related to SOD enzyme activity: the greater the activity, the lower the absorbance.
