Description
The Cholesterol/Cholesterol ester quantitation kit can be used to detect free cholesterol, cholesteryl esters, or both through quantitative analysis of absorbance (colorimetric) and fluorescence (fluorometric) measurements. Most of the cholesterol in blood exists in the form of cholesteryl esters which are hydrolyzed to free cholesterol by cholesterol esterase. Cholesterol is then oxidized by cholesterol oxidase to produce H2O2 as a byproduct, which reacts with the cholesterol probe to produce color (maximum absorbance 570 nm) and fluorescence (Ex/Em = 535/590 nm). The assay detects free cholesterol in the absence of cholesterol esterase and total cholesterol (cholesterol and cholesteryl esters) in the presence of cholesterol esterase. The amount of cholesteryl ester is determined by taking the difference between the value of total cholesterol and the value of free cholesterol.
